Product Specific Information
The SB/199 monoclonal antibody reacts with mouse CD127, the high-affinity α subunit of the mouse IL-7 receptor.
IL-7 receptor α chain is expressed by immature B cells in the bone marrow, double-negative (CD4-/CD8-), single-positive (CD4+ and CD8+) thymocytes, but not double-positive (CD4+/CD8+) thymocytes. Mature peripheral T cells express low levels of CD127.
Binding of SB/199 blocks the binding of A7R34, another antibody recognizing mouse CD127.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤1 µg/test (100 µL final volume). Optimal titration recommended.
Super Bright 702 is a tandem dye excited with the violet laser (405 nm) and emits at 702 nm. Recommended filter: 710/50 bandpass.
Use Super Bright Complete Staining Buffer (SB-4401) when combining multiple Super Bright dyes to minimize non-specific polymer interactions.
Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (00-8222) or 1-step Fix/Lyse Solution (00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information
CD127 (Interleukin-7 receptor α) is a glycoprotein involved in lymphopoiesis regulation.
It forms a heterodimer with the γ chain (shared with IL-2 receptor) to activate IL7R signaling.
CD127 promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature CD4+/CD8+ T cells.
Dysfunction is associated with severe combined immunodeficiency (SCID) and T cell negative/B cell negative/NK positive SCID.
