Product Specific Information

The ACK2 monoclonal antibody reacts with mouse CD117 (c-Kit receptor), also known as Steel factor receptor and stem cell factor receptor. This 145 kDa tyrosine kinase receptor is expressed by hematopoietic progenitor cells, mast cells, and plays a key role in cell proliferation and differentiation. ACK2 has been reported as a blocking antibody.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometry on mouse bone marrow cells (≤0.5 µg/test). A test is defined as staining a cell sample in 100 µL total volume. Cell number can range from 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Super Bright 702 Characteristics:

• Tandem dye excited by 405 nm violet laser, emits at 702 nm.
• Recommended filter: 710/50 bandpass.
• Use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize polymer interactions when using multiple Super Bright conjugates.
• Sensitive to photo-induced oxidation—protect from light.

Fixation Guidance:
Samples may be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

KIT (c-KIT) is a proto-oncogene encoding a type 3 transmembrane receptor for mast cell growth factor (MGF, stem cell factor). It regulates proliferation and activation of hematopoietic and non-hematopoietic cells. Mutations in KIT are linked to gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism.

Upon SCF binding, c-Kit dimerizes and autophosphorylates tyrosine residues (e.g., Tyr703), enabling Grb2 docking and Ras/ERK pathway activation. KIT signaling also activates PI3K, PLC-γ, and JAK/STAT pathways, essential for hematopoiesis, stem cell maintenance, and gametogenesis.
