Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8αβ) or homodimer (CD8αα). It is found on most thymocytes and subsets of mature T cells and NK cells. CD8 binds to MHC class I and, through association with p56lck, plays a key role in T-cell development and activation.

Preliminary testing shows that OKT8, RPA-T8, and HIT8a clones do not compete for binding to human peripheral blood leukocytes, suggesting distinct epitopes.

Applications Tested: Flow cytometric analysis of normal human peripheral blood cells. Recommended ≤0.25 µg/test in 100 µL final volume. Cell number may vary (10⁵–10⁸ cells/test). Optimize titration for best results.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor with the T cell receptor (TCR) for MHC class I molecules and is primarily expressed on cytotoxic T cells as a CD8αβ heterodimer. CD8α also forms homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells.

Upon MHC-I/peptide complex ligation, CD8 triggers recruitment of Lck kinase, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are crucial for pathogen and tumor cell clearance. Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
