The monoclonal antibody C8/144B recognizes the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8α/β) or homodimer (CD8α/α). CD8a is expressed on most thymocytes, subsets of mature T cells, and NK cells. It binds to MHC class I and, through association with protein tyrosine kinase p56lck, contributes to T cell development and activation.

Applications Reported:
This antibody is suitable for immunohistochemical staining of formalin-fixed, paraffin-embedded (FFPE) tissue sections.

Applications Tested:
Tested on FFPE human tonsil tissue using IHC Antigen Retrieval Solution - High pH (Product # 00-4956-58). Recommended concentration ≤ 5 µg/mL. Optimal dilution should be determined experimentally.

Target Information

Cluster of Differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family involved in signal transduction and T cell differentiation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer (CD8α/β), functioning as a co-receptor with TCR for MHC class I molecules. CD8α is essential for MHC-I binding and is also expressed as a homodimer (CD8α/α) on subsets of T cells, NK cells, monocytes, and dendritic cells.

Upon MHC-I/peptide complex ligation, CD8 recruits the tyrosine kinase Lck, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are critical for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
