Intact IgG appears on a non-reducing gel as a ~150 kDa band and upon reduction generates a ~25 kDa light chain band and a ~50 kDa heavy chain.

Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems by cloning specific antibody DNA sequences from immunoreactive rabbits. Individual clones are screened to select optimal candidates for production.

Target Information

IkB-alpha is a 40 kDa protein that inhibits NF-kappaB activity through protein-protein interaction in the cytosol. This interaction masks the nuclear localization sequence of NF-kappaB, preventing its translocation to the nucleus.

Various stimuli such as TNF-alpha, IL-1 beta, chemokines, PMA, growth factors, LPS, UV irradiation, viral infection, and other stresses can activate NF-kappaB by promoting IkB-alpha degradation.

In humans, the gene is located on chromosome 14q. Activation of NF-kappaB requires phosphorylation of IkB on specific serine residues, leading to its degradation. IkB kinase alpha (IKK alpha, also known as CHUK) interacts with and phosphorylates IkB-alpha at Ser32 and Ser36, triggering degradation.

IKK alpha plays a critical role in NF-kappaB activation in response to proinflammatory cytokines. Phosphorylation of IkB by IKK alpha is stimulated by NFkB-inducing kinase (NIK), a central regulator of NF-kappaB activation in response to TNF and IL-1. The functional IKK complex includes IKK alpha, IKK beta, and IKK gamma, each contributing to IkB phosphorylation.
