This antibody detects endogenous levels of PAK3 only when phosphorylated at serine 154.

Target Information

PAK proteins are critical effectors that link Rho GTPases to cytoskeleton reorganization and nuclear signaling. PAK proteins, a family of serine/threonine p21-activating kinases, serve as targets for the small GTP binding proteins Cdc42 and RAC and have been implicated in a wide range of biological activities. The α, β, and γ PAK isoforms complex specifically with Rac1 and Cdc42 in their active GTP-bound state, inhibiting their intrinsic GTPase activity leading to their autophosphorylation. There are eight sites of autophosphorylation on γPAK, including Ser19, Ser141, and Thr402, and phosphorylation of Ser141 and Thr402 is correlated with γPAK activation. Once phosphorylated and their affinity for Rac/Cdc42 reduced, the PAK isoforms dissociate from the complex to seek downstream substrates.
