Product Specific Information

This antibody detects endogenous levels of ROCK1 only when phosphorylated at Tyr913.

Target Information

ROCK1 is a serine/threonine kinase that regulates cell differentiation and migration. The Rho/ROCK pathway is involved in the progression of various human cancers. ROCK1 has been identified as a Caspase-3 substrate. It contributes to myosin light chain (MLC) activation through direct phosphorylation and inhibition of MLC phosphatase, leading to increased cellular contractility required for apoptotic membrane blebbing.

ROCK1 consists of an amino-terminal kinase domain and a cysteine/histidine-rich C-terminal domain within a pleckstrin-homology region, connected by a variable region containing the Rho-binding domain. During apoptosis, ROCK1 is cleaved by caspase-3 at a conserved DETD1113/G sequence, removing its inhibitory domain and resulting in constitutive kinase activity. This cleavage induces MLC phosphorylation and membrane blebbing, key events in apoptosis execution.
