Product Specific Information

Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
Suggested positive control: colon carcinoma or rectal carcinoma.


Target Information

E-Cadherin (epithelial cadherin) is a classical cadherin belonging to the calcium-dependent adhesion protein superfamily.
It is a cell-cell adhesion glycoprotein consisting of five extracellular cadherin repeats, a transmembrane region, and a conserved cytoplasmic tail.

Mutations in the CDH1 gene are associated with gastric, breast, colorectal, thyroid, and ovarian cancers.
Loss of E-cadherin function contributes to cancer progression by increasing proliferation, invasion, and metastasis.
The ectodomain mediates bacterial adhesion to mammalian cells, while the cytoplasmic domain is required for internalization.

E-cadherin plays a crucial role in tissue architecture and integrity maintenance.
In humans, it is encoded by the CDH1 gene on chromosome 16.
Reduced or lost expression of E-cadherin in carcinomas correlates with increased invasion and metastasis potential.
