• Heat-mediated antigen retrieval is recommended prior to staining, using an EDTA buffer for 10 minutes followed by cooling at room temperature for 20 minutes.
• Following antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
• A suggested positive control is HeLa cell lysate.

Target Information

CDK2/cyclinO is a member of the cyclin-dependent protein kinase family complexed to CyclinO. CyclinO, a specific protein isoform name for the UNG gene, is localized exclusively in the nucleus and exhibits uracil-DNA glycosylase activity. CDK2/cyclinO protein levels increase during the G1 phase and are turned over during the cell cycle. Cyclin A–cdk2 levels peak at the G1/S transition, and both are associated with DNA in the initiation complex during replication. Rb protein acts as a substrate for cdk2–cyclin E and/or cdk2–cyclin A in vivo. Cdk2 is activated and localized to the nucleus during late G1-, S-, and G2-phase.
