Product Specific Information

The monoclonal antibody 10F7MN recognizes human glycophorin A (CD235a), detecting both M and N alleles.
Glycophorin A is a 151 amino acid sialoglycoprotein found on erythrocyte and erythroid progenitor cell membranes (~500,000 copies per cell).
The gene resides on chromosome 4 with two allelic forms (M and N), differing in two amino acids:

  • M group: Ser1, Gly5
  • N group: Leu1, Glu5

Exposure to toxins may cause allele mutation or loss, leading to phenotypic changes. Studies suggest correlations between genotype/phenotype and predisposition to cancer and heart disease.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells.
Use at 5 µL (0.8 µg) per test (100 µL final volume). Cell number range: 10^5–10^8 cells/test.

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

This NovaFluor conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance.
Add 5 µL per sample to antibody cocktails before combining with cells.
For single-color controls: use 5 µL per 100 µL cell sample (10^3–10^8 cells).

NovaFluor conjugates are based on Phiton™ technology, utilizing engineered nucleic acid dye structures that minimize spillover and spread.

Target Information

CD235a (Glycophorin A, GPA) is a single-pass membrane sialoglycoprotein expressed on mature erythrocytes and erythroid precursor cells.
It carries blood group M and N specificities and provides a mucin-like barrier to prevent red cell aggregation.
CD235a also acts as a receptor for Sandei virus, parvovirus, and Streptococcus adhesin (Hsa).
