Product Specific Information

The M1/69 monoclonal antibody reacts with mouse CD24 (Heat Stable Antigen, HSA), a 35–50 kDa molecule anchored via phosphatidylinositol. It is expressed by erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage cells. CD24 shows variable glycosylation, leading to heterogeneity in molecular mass across cell types. Expression levels can help identify stages of B lymphopoiesis in mouse bone marrow. P-selectin (CD62P) has been reported to bind CD24.

Each product includes:

Applications Tested: Mouse splenocytes and thymocytes (≤0.8 µg/test, 100 µL final volume)

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
Recommended usage:

• Add 5 µL per sample before combining with cells.
• For single-color controls, add 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, enabling engineered fluorescent signatures with minimized spillover and spread.

Internal testing shows that NovaFluor Yellow 590 may non-specifically stain B cells in SJL mice. No non-specific staining observed in BALB/c or C57BL/6 strains.

Target Information

CD24 is a GPI-anchored glycoprotein expressed at multiple stages of B-cell development—from pro-B cells in bone marrow to mature B cells. Plasma cells show low or no expression. CD24 is also found in most B-lineage acute lymphoblastic leukemias, B-cell CCLs, and B-cell non-Hodgkin’s lymphomas. It may regulate B-cell proliferation, maturation, and autoimmunity control.
