Product Specific Information

The M1/70 monoclonal antibody reacts with mouse CD11b, a 165–170 kDa integrin alphaM. CD11b associates with CD18 to form Mac-1 (αMβ2 integrin), binding to CD54 (ICAM-1), C3bi, and fibrinogen. It is expressed by macrophages, NK cells, granulocytes, activated lymphocytes, and mouse B-1 cells. This antibody is cross-reactive to human CD11b and can detect the antigen on human peripheral blood monocytes, granulocytes, and NK cells.

Applications Tested: Mouse splenocytes and bone marrow cells (≤ 0.4 µg/test)

A test is defined as staining a cell sample in 100 µL volume, with cell numbers ranging from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.

Compatibility Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Recommended alternatives: Invitrogen LIVE/DEAD Fixable Dead Cell stains.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) should be used for optimal staining:

• Add 5 µL per sample before combining antibody cocktails with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescent signatures minimizing spillover and spread.

Target Information

CD11b (integrin alpha-M, ITGAM) is a 165 kDa adhesion molecule that associates with CD18 to form the Mac-1 (αMβ2 integrin) complex. It functions as a receptor for ICAMs, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X. CD11b is expressed on monocytes/macrophages, granulocytes, NK cells, dendritic cells, and microglia. Expression levels vary by tissue and increase during monocyte maturation. It serves as a microglial marker in nervous tissue and is implicated in systemic lupus erythematosus and ITGAM-related susceptibility disorders.
