Product Specific Information

The M1/70 monoclonal antibody reacts with mouse CD11b, a 165–170 kDa integrin alphaM. CD11b associates with CD18 to form the alphaMbeta2 integrin (Mac-1) and binds CD54 (ICAM-1), C3bi, and fibrinogen.
Mac-1 is expressed by macrophages, NK cells, granulocytes, activated lymphocytes, and mouse B-1 cells. M1/70 is cross-reactive to human CD11b, useful for detecting this antigen on human peripheral blood monocytes, granulocytes, and some NK cells.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse splenocytes and bone marrow cells (≤ 0.4 µg/test).
A test is defined as staining a cell sample in 100 µL volume; cell count may range from 10⁵–10⁸ cells/test.
Careful titration is recommended for optimal performance.

Compatibility Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD11b (integrin alpha-M, ITGAM) is a 165 kDa adhesion molecule that forms a heterodimer with CD18, known as Mac-1 or CR3.
It is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, subsets of NK and dendritic cells, and microglia.
Functions as receptor for ICAMs, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, mediating cell-cell and cell-matrix interactions.
Expression levels vary by tissue and cell type; used as a microglial marker.
Associated with systemic lupus erythematosus and ITGAM-related susceptibility disorders.
