Product Specific Information

The M1/69 monoclonal antibody reacts with the mouse CD24 molecule, also known as Heat Stable Antigen (HSA). This 35–50 kDa GPI-anchored molecule is expressed on erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage cells. CD24 expression varies due to glycosylation heterogeneity and is used to identify B lymphopoiesis stages in mouse bone marrow. It is also known to bind P-selectin (CD62P).

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes and thymocytes at ≤ 0.8 µg/test in 100 µL final volume. Cell number may range from 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, engineered for optimized fluorescence with minimal spillover and spread.

Note: Internal testing shows NovaFluor Yellow 660 may non-specifically stain B cells in SJL mice. No non-specific staining observed in BALB/c or C57BL/6 mice.

Target Information

CD24 is a GPI-anchored glycoprotein expressed at multiple stages of B-cell development, from pro-B cells to mature B cells. Expression is low or absent in plasma cells. CD24 is also found in B-lineage acute lymphoblastic leukemias, B-cell CCLs, and B-cell non-Hodgkin’s lymphomas. It may regulate B-cell proliferation, maturation, and autoimmunity control.
