Mutations, rearrangements, and amplifications in the ALK gene are associated with various tumors including anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer.
Chromosomal rearrangement commonly results in a fusion gene between ALK and the nucleophosmin (NPM) gene, producing an oncogenic NPM-ALK fusion protein.
ALK encodes a receptor protein-tyrosine kinase with a transmembrane and extracellular domain, showing high sequence similarity to LTK.
