Target Information
FOXO1 (FKHR, Forkhead Box O1) is a transcription factor belonging to the forkhead family. It can act as a coactivator or corepressor of nuclear receptor activity through the LXXLL motif in its C-terminal region. FOXO1 interaction with PAX3 is linked to alveolar rhabdomyosarcoma. It plays key roles in autophagy, myogenic differentiation, and tumor suppression.
FOXO1 is a major target of insulin signaling, regulating metabolic homeostasis and oxidative stress response. It binds to insulin response elements (IRE) and Daf-16 family binding elements (DBE) to control redox balance, osteoblast function, and bone mass. Additionally, FOXO1 influences glucose metabolism by regulating genes such as IGFBP1, G6PC, and PPCK1 in hepatocytes.
