Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Rat TNF-alpha. Anti-Rat TNF-alpha-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect Rat TNF-alpha by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Rat TNF-alpha (500-P72) as a capture antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant Rat TNF-alpha.

Western Blot:
To detect Rat TNF-alpha by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Rat TNF-alpha is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha plays roles in immune regulation, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It forms multimers (dimers, trimers, or pentamers) and appears as a single 17 kDa band on SDS-PAGE under non-reducing conditions. Knockout studies suggest neuroprotective functions and tumor necrosis activity. TNF alpha is involved in immune responses to various infections and tumor necrosis, and has been implicated in autoimmune diseases, insulin resistance, and cancer.
