Preparation
Produced from sera of rabbits immunized with highly pure Recombinant Rat TNF-alpha. Anti-Rat TNF-alpha-specific antibody was purified by affinity chromatography employing an immobilized Rat TNF-alpha matrix.
Sandwich ELISA
To detect Rat TNF-alpha by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Rat TNF-alpha (500-P72Bt) as a detection antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Rat TNF-alpha.
Western Blot
To detect Rat TNF-alpha by Western Blot analysis, use at a concentration of 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Rat TNF-alpha is 1.5–3.0 ng/lane under reducing or non-reducing conditions.
Target Information
TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cell activity, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It exists as multimers but shows a single 17 kDa band under non-reducing SDS-PAGE conditions. Knockout studies suggest neuroprotective roles, and it can induce tumor necrosis in tumor-bearing mice. TNF alpha is involved in immune responses to infections and in the necrosis of certain tumors, and has been implicated in autoimmune diseases, insulin resistance, and cancer.
