This antibody detects endogenous levels of Cyclin D1 only when phosphorylated at Threonine 286.


Target Information

Cyclin D1 (also known as PRAD-1 or bcl-1) is a key regulator of the cell cycle that functions with CDK4 and/or CDK6 to phosphorylate the Rb protein. It acts as a proto-oncogene and is overexpressed in various human cancers, including mantle cell lymphoma (MCL). Cyclin D1 regulates protein phosphorylation, epithelial cell proliferation, and adipocyte differentiation. The CCND1 gene is located on chromosome 11 and encodes a cytoplasmic and nuclear protein synthesized during G1 phase. Cyclin D1 forms complexes with CDK4 or CDK6, promoting G1/S transition by inactivating Rb and modulating CDK inhibitors (p21Cip1, p27Kip1). Phosphorylation triggers its ubiquitin-mediated degradation, leading to G1/S arrest. Due to its critical role in cell cycle control, Cyclin D1 is a valuable marker and research target in oncology and cell biology.
