Product Specific Information

The eBio1D3 (1D3) monoclonal antibody reacts with mouse CD19, a 95 kDa transmembrane glycoprotein expressed by B cells during all developmental stages except plasma cells. Follicular dendritic cells also express CD19. CD19 forms a multimolecular complex with CD21, CD81, and MHC class II, associating with the BCR to mediate signaling events such as tyrosine phosphorylation, calcium flux, and B cell proliferation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤ 0.8 µg/test (100 µL final volume). Cell count: 10⁵–10⁸ cells/test. Optimize titration for best results.

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer Usage:
Use 5 µL per sample (regardless of NovaFluor count). For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells). Add buffer to antibody cocktails before combining with cells. Required for optimal staining.

Technology:
NovaFluor conjugates utilize Phiton™ technology based on nucleic acid dye structures for engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD19, a member of the immunoglobulin superfamily, has two Ig-like domains and is expressed on all peripheral B cells. It is also found on certain myeloid leukemia cells. CD19 acts as a signal-amplifying coreceptor for the B cell receptor (BCR) and regulates normal and malignant B cell proliferation. Mutations cause hypogammaglobulinemia, while overexpression leads to B cell hyperactivity.
