Product Specific Information

The eBio1D3 (1D3) monoclonal antibody reacts with mouse CD19, a 95 kDa transmembrane glycoprotein expressed on B cells during all developmental stages except terminally differentiated plasma cells. Follicular dendritic cells also express CD19. CD19 forms a complex with CD21, CD81, and MHC class II, associating with the BCR to mediate signaling that induces tyrosine phosphorylation, calcium flux, and B cell proliferation.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.8 µg/test (100 µL final volume). Optimize titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains, expressed on 100% of peripheral B cells and some myeloid leukemia cells. It is a key regulator of B cell proliferation and differentiation, serving as a co-receptor with the BCR to enhance antigen sensitivity. CD19 mutations cause hypogammaglobulinemia, while overexpression leads to B cell hyperactivity.
