Product Specific Information

The OKT4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface glycoprotein expressed by most thymocytes, mature T helper cells, and at low levels on monocytes. CD4 acts as a receptor for HIV. The OKT4 antibody recognizes a distinct epitope from RPA-T4 and does not cross-block binding.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Validated on normal human peripheral blood cells at ≤5 µL (1.0 µg) per 100 µL reaction (10⁵–10⁸ cells/test). Optimize titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails before mixing with cells.
  • For single-color controls: 5 µL buffer per 100 µL of cell sample (10³–10⁸ cells).

Technology: NovaFluor conjugates utilize Phiton™ technology based on nucleic acid dye structures engineered for optimized fluorescence with reduced spectral spillover and spread.

Target Information

CD4 is a co-receptor for MHC class II molecules and HIV, primarily expressed on T helper cells, and also on monocytes, macrophages, and dendritic cells. In tissues, CD4 is present in thymus, lymph nodes, tonsils, spleen, and certain brain and gut regions. It plays a key role in T-cell activation and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
