Product Specific Information

The OKT4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface glycoprotein expressed by thymocytes, T-helper cells, and at low levels on monocytes. CD4 functions as a receptor for HIV. The OKT4 antibody recognizes a distinct epitope from RPA-T4 and does not cross-block binding.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤ 5 µL (1.0 µg) per test in 100 µL final volume. Cell count range: 10⁵–10⁸ cells/test. Optimize by titration for best results.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates use Phiton technology with nucleic acid dye structures engineered for optimized fluorescent signatures, minimizing spillover and spread.

Target Information

CD4 antigen participates in MHC class II recognition and acts as a co-receptor for HIV. It is primarily expressed in T-helper cells but also in monocytes, macrophages, and dendritic cells. CD4 expression is found in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. It plays a role in T-cell activation via association with the T-cell receptor complex and protein tyrosine kinase Lck, and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
