• Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Placental Alkaline Phosphatase (PLAP) plays an important role in regulating inflammatory disease processes. There are four distinct alkaline phosphatases: intestinal, placental, placental-like, and liver/bone/kidney types. PLAP is a membrane-bound isoenzyme (Regan and Nagao type) expressed in the placenta during the third trimester. It is useful in identifying testicular germ cell tumors. PLAP-positive somatic cell tumors also express epithelial membrane antigen (EMA). Although PLAP is proposed to function in matrix mineralization, mice lacking functional enzyme exhibit normal skeletal development. PLAP has been linked to hypophosphatasia, a disorder characterized by hypercalcemia and skeletal defects, with severity depending on mutation type.
