Product Specific Information
The J105 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the CD28 immunoglobulin superfamily. PD-1 plays a key role in peripheral tolerance and autoimmune disease and is expressed predominantly on activated T and B lymphocytes. Its ligands PD-L1 (B7-H1) and PD-L2 (B7-DC) show overlapping functions and are expressed on normal and activated antigen-presenting cells.
Costaining experiments indicate eBioJ105 recognizes a different epitope than MIH4 (Product #11-9969).
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry on normal human peripheral blood cells at 5 µL (0.5 µg)/test (100 µL final volume, 10⁵–10⁸ cells/test).
Brilliant Violet™ 711 (BV711):
- Tandem dye emitting at 713 nm for use with 405 nm violet laser.
- Use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific polymer interactions.
- Sensitive to light; protect vial and stained samples.
- Fixation compatibility: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) up to 3 days at 4°C in dark.
- Not compatible with methanol-based fixation.

Target Information
T-cell activation is regulated by PD-1, which delivers inhibitory signals upon binding to its ligands PD-L1 and PD-L2 on antigen-presenting cells. PD-1 deficiency leads to hyperactivated immune responses and autoimmune disease. Despite its predicted molecular weight, PD-1 often migrates at higher molecular weight in SDS-PAGE.
