Product Specific Information

This 3DS223H monoclonal antibody recognizes human CD223 (LAG-3), a 70-kDa surface glycoprotein of the Ig superfamily with homology to CD4. LAG-3 binds to MHC class II with higher affinity than CD4 and negatively regulates T cell activation and proliferation. It is expressed on activated T cells (including regulatory T cells) and NK cells, with higher levels on CD8+ T cells.

• Tested on stimulated normal human peripheral blood cells
• Recommended use: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Fluorochrome Information:
Brilliant Violet™ 711 (BV711) emits at 713 nm and is optimized for violet (405 nm) laser-equipped cytometers. Protect from light due to photo-induced oxidation sensitivity.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Fixation Compatibility:
Samples may be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark. BV711 is not compatible with methanol-based fixation.

Target Information

LAG-3 (Lymphocyte Activation Gene 3) is a 70-kDa surface glycoprotein related to CD4. It binds MHC class II with high affinity and regulates T cell activation and homeostasis. Expressed on activated T cells and NK cells, with higher expression in CD8+ T cells. Coexpression with CD49b identifies Type 1 regulatory T cells (Tr1).
