Product Specific Information

The 53-2.1 monoclonal antibody reacts with mouse CD90.2 (Thy-1.2), a GPI-linked membrane molecule expressed by thymocytes, mature T cells, and neurons in CD90.2-expressing mouse strains (BALB/c, CBA, C3H, C57BL/6, C58, SJL, etc.). Strains expressing CD90.1 (PL, AKR) do not stain with 53-2.1.

CD90 is involved in regulation of adhesion and signal transduction by T cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.125 µg/test (100 µL final volume). Optimal titration recommended.

Fluorochrome Information:
Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Recommended filter: 660/20 bandpass. Ensure instrument compatibility.

Staining Panel Considerations:
When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect vial and stained samples from light due to photo-induced oxidation.

Fixation:
Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal impact on brightness and FRET efficiency.

Target Information

CD90.2 (Thy-1) is a GPI-linked glycoprotein of the immunoglobulin superfamily. It is expressed in murine T cells, thymocytes, neural cells, Kupffer cells, and fibroblasts. CD90.2 is involved in cell-cell interactions during synaptogenesis and other neural processes. It is an 18 kDa plasma membrane antigen highly expressed in connective tissue, fibroblast and stromal cell lines, and tumor endothelial cells. In mice, CD90.2 is expressed on all thymocytes and peripheral T lymphocytes.
