Description:
The Sa2-8 monoclonal antibody reacts with mouse CD14, a 53–55 kDa GPI-linked glycoprotein. CD14 is a receptor for LPS–LBP complexes and associates with Toll-Like Receptor 4 (TLR4), participating in signaling and cellular responses to bacterial LPS. In mice, CD14 is expressed on macrophage surfaces and can also be found in serum as a secreted form. Sa2-8 exhibits weak antagonistic activity in NF-κB activation or TNF-α production upon LPS stimulation.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse thioglycolate-elicited peritoneal exudate cells. Recommended use ≤0.5 µg/test (100 µL final volume; 10⁵–10⁸ cells/test). Optimal titration is recommended for best performance.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a lipopolysaccharide (LPS) receptor and exists in both membrane-bound and soluble forms. CD14 binds LPS via LBP, contributing to immune recognition and inflammatory responses. It also binds apoptotic cells and is involved in allergic, inflammatory, and infectious processes. Alternative splicing results in multiple transcript variants encoding the same isoform. CD14 dysfunction has been linked to mycobacterium chelonae infection and croup.
