Product Specific Information

The monoclonal antibody 2D3 reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein involved in adhesion via interactions with SIRP (signal regulator protein) and integrins CD51/CD61 and CD41/CD61. It mediates bi-directional signaling affecting neural synaptic activity and macrophage phagocytosis. CD47 serves as a receptor for thrombospondin and is expressed on most hematopoietic and non-hematopoietic cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.6 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining. Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

Target Information

CD47 (Integrin-associated protein, IAP) is a ubiquitously expressed transmembrane glycoprotein interacting with integrins and modulating signaling pathways such as G protein activation. It regulates intracellular calcium increase upon adhesion to the extracellular matrix and acts as a receptor for thrombospondin. CD47 serves as a ligand for CD172a (SIRPα), inhibiting macrophage phagocytosis of CD47-positive cells. It also influences cell migration, B/T cell activation, chondrocyte mechanotransduction, and neuronal development.
