Product Specific Information

The SN3 monoclonal antibody reacts with the human CD24 molecule, also known as Heat Stable Antigen (HSA). This 35–50 kDa molecule is anchored in the plasma membrane via phosphatidylinositol and is expressed by erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage. P-selectin (CD62P) has been reported to bind to CD24.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage is 5 µL (0.2 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample to antibody cocktails/master mixes prior to combining with cells.
• For single-color controls, use 5 µL of CellBlox Blocking Buffer per 100 µL of cell sample containing 10³–10⁸ cells.

NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures that enable engineered fluorescent signatures with optimized spillover and spread characteristics.

Target Information

CD24 is a GPI-anchored glycoprotein expressed at multiple stages of B-cell development, from pro-B cells in bone marrow to mature surface Ig-positive B cells. Plasma cell expression is low or negative. CD24 is also expressed on most B-lineage acute lymphoblastic leukemias, B-cell CCLs, and B-cell non-Hodgkin’s lymphomas. It may play a role in B-cell proliferation, maturation, and autoimmunity regulation.
