Product Specific Information

The eBioA1 monoclonal antibody reacts with human CD39, also known as ectonucleoside triphosphate diphosphohydrolase 1 (ENTPD1) or NTPDase. CD39 is an integral membrane protein with two transmembrane domains and forms a homotetramer. It is the most prominent ectoenzyme of the immune system, functioning to remove toxic extracellular ATP by converting it to ADP or AMP. CD39 works together with CD73 to hydrolyze ATP and is well characterized on Langerhans cells.

Expression of CD39 was originally identified on activated lymphocytes and is also found on subsets of T cells, B cells, dendritic cells, with weak expression on monocytes and granulocytes. CD39 and CD73 are expressed on regulatory T cells (Treg), facilitating their entry into inflamed areas with high ATP levels. Expression on Foxp3+CD4+ cells ranges from 25–45%.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated for flow cytometry on normal human peripheral blood cells. Recommended use: 5 µL (0.8 µg) per test. A test is defined as staining a cell sample in a final volume of 100 µL (10⁵–10⁸ cells/test). Antibody should be titrated for optimal assay performance.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Updated conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01). This improved formulation is required for optimal staining with NovaFluor conjugates.

  • Add 5 µL per sample (regardless of number of NovaFluors) before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology based on novel nucleic acid dye structures, engineered for optimized fluorescent signatures with minimal spillover and spread.

Target Information

CD39 is a 70–100 kDa cell surface molecule expressed on peripheral blood B cells, monocytes, T cell clones, and weakly on granulocytes. It has intrinsic ecto-ATPase activity. Expression is induced on T cells and increased on B cells as a late activation antigen.
