Product Specific Information
The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, mature T cell subpopulations, and dendritic cells. CD4 binds to MHC class II molecules on antigen-presenting cells and plays key roles in T cell development and activation. In T cells, CD4 associates with protein tyrosine kinase p56lck via its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.
Applications Reported: Flow cytometric analysis.
Applications Tested: Validated in flow cytometry using mouse splenocytes at ≤0.125 µg/test (100 µL final volume; 10⁵–10⁸ cells/test). Optimal titration recommended.
Fluorochrome Information:
eFluor® 506 is excited by the 405 nm violet laser and emits at 506 nm. Recommended detection filter: 510/20 band pass or equivalent. Verify instrument compatibility.
Fixation:
Samples may be stored up to 3 days at 4°C in the dark using:
- IC Fixation Buffer (Product #00-8222-49)
- 1-step Fix/Lyse Solution (Product #00-5333-54)
Clone-specific performance post-fixation should be empirically confirmed.

Target Information
CD4 antigen is a co-receptor for MHC class II molecules and HIV. It is mainly expressed on T helper cell subsets, as well as monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and certain brain and gut regions. CD4 aids early T-cell activation via association with TCR complex and Lck kinase, and may also mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
