Product Specific Information

The RPA-T4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface receptor expressed by most thymocytes, a subset of mature T cells (T-helper cells), and at low levels on monocytes. CD4 serves as a receptor for HIV. The RPA-T4 antibody blocks HIV binding and mixed lymphocyte reactions. It recognizes a different epitope than the OKT4 monoclonal antibody, and the two do not cross-block each other.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-titrated and validated for flow cytometry of normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per 100 µL test volume, with 10⁵–10⁸ cells/test.
  • Fluorochrome Information: eFluor® 506 is excited by the 405 nm violet laser and emits at 506 nm. Recommended detection filter: 510/20 band pass or equivalent.
  • Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD4 is involved in the recognition of MHC class II molecules and acts as a co-receptor for HIV. It is primarily expressed on T-helper cells, and also found on monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut. CD4 mediates early T-cell activation via association with the T-cell receptor complex and protein tyrosine kinase Lck. It may also contribute to neuronal damage in infectious and immune-mediated CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
