Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta subunits, is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells.

Crosslinking of TCR via immobilized UCHT1 initiates intracellular signaling leading to activation and proliferation.

• Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells
• Recommended Use: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Target Information

The CD3 complex is crucial for transmitting antigen-recognition signals in T cells and regulating TCR surface expression. It consists of subunits CD3γ, CD3δ, CD3ε, and CD3ζ, encoded on chromosome 11. These subunits have cytoplasmic tails associated with signaling molecules through tyrosine-based motifs.

CD3 is expressed on:

• 68–82% of normal peripheral blood lymphocytes
• 65–85% of thymocytes
• Purkinje cells in the cerebellum

Not expressed on B or NK cells. CD3 defects are linked to immunodeficiency and altered T cell percentages in autoimmune diseases.
