Description:
The WM-53 monoclonal antibody reacts with human CD33 (GP67, P67), a 67 kDa type I transmembrane glycoprotein belonging to the Siglec family. CD33 is expressed in the hematopoietic compartment, including monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.

Applications Reported:
The WM-53 (WM53) antibody is reported for use in:

• Flow cytometric analysis
• Immunoprecipitation
• Immunocytochemistry staining

Applications Tested:
Tested by flow cytometric analysis of lysed whole blood. Recommended ≤1 µg per test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Optimal titration recommended.

Target Information

CD33 is a transmembrane protein in the Siglec family with ITIM motifs that recruit SHP-1 and SHP-2 for inhibitory signaling. It binds sialic acid residues on cell surface glycans and serves as a therapeutic target for acute myeloid leukemia. CD33 is expressed on myelomonocytic lineage cells, granulocyte and macrophage precursors, and peripheral monocytes, but not on pluripotent stem cells. It helps distinguish myelogenous leukemia cells from lymphoid or erythroid leukemias. CD33 dysfunction has been associated with gallbladder lymphoma and extracutaneous mastocytoma.
