The HIM3-4 monoclonal antibody reacts with human CD33, a 67 kDa member of the sialoadhesion family. CD33 is expressed by myelomonocytic precursors, monocytes, mast cells, and granulocytes, but not by hematopoietic stem cells or lymphocytes. CD33 plays a role in sialic-acid dependent cell adhesion.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on normal human peripheral blood cells at ≤1 µg/test (100 µL final volume). Optimal titration is recommended for best performance.

Target Information
CD33 is a transmembrane protein of the Siglec family containing immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit SHP-1 and SHP-2 phosphatases. It mediates sialic acid–dependent interactions and is expressed on myelomonocytic lineage cells, granulocyte and macrophage precursors, and peripheral monocytes. CD33 serves as a diagnostic and therapeutic target in acute myeloid leukemia and helps distinguish myelogenous leukemia cells from lymphoid or erythroid types. Dysregulation of CD33 is associated with gallbladder lymphoma and extracutaneous mastocytoma.
