Product Specific Information
The GL1 monoclonal antibody reacts with mouse CD86 (B7-2), an ~80 kDa surface receptor and member of the B7 family of costimulatory molecules. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells, and is upregulated through stimuli such as BCR complex, CD40, and cytokine receptors. It is also expressed by activated mouse T cells and thioglycolate-elicited peritoneal cells.
CD86 interacts with T cell surface molecules CD28 and CD152 (CTLA-4), playing a critical role in T-B cell crosstalk, T cell costimulation, and antibody production. Its rapid upregulation upon stimulation indicates a key role during the primary immune response.
Applications Reported: Flow cytometry, immunoprecipitation, frozen tissue staining, and blocking studies (functional assays should use Functional Grade purified GL1, Product #16-0862).
Applications Tested: Tested by flow cytometric analysis of resting and activated mouse splenocytes. Recommended ≤0.5 µg/test (100 µL volume). Optimal titration is advised for specific assay conditions.

Target Information
CD86 is one of two ligands (along with CD80) for CTLA4 and CD28, functioning as a costimulatory molecule in T-cell activation. Insufficient co-stimulation via CD86/CD80 can lead to immune tolerance. CD86 is a type I membrane protein of the immunoglobulin superfamily, expressed on antigen-presenting cells. It binds CD28 (activating signal) and CTLA-4 (inhibitory signal) on T cells, regulating immune responses. Alternative splicing generates multiple isoforms. CD86 dysfunction is associated with diseases such as gallbladder squamous cell carcinoma and myocarditis.
