The monoclonal antibody R139 recognizes human CD93 (C1qRp), a glycoprotein that binds to complement protein C1q, mannose binding lectin, and pulmonary surfactant protein A. CD93 plays a role in the clearance of apoptotic cells and is mainly expressed on myeloid cells, with higher levels on monocytes than on neutrophils, eosinophils, platelets, and endothelial cells. Expression on dendritic cells decreases upon maturation. CD93 also defines an early bone marrow stem cell population of hematopoietic and hepatic precursors.

The R139 antibody blocks C1q-mediated enhancement of phagocytosis and can detect soluble CD93 by ELISA when used in combination with R3 antibody. The epitope for R139 is located in the EGF domains.

Applications Reported: Flow cytometry, immunoprecipitation, immunoblotting (WB) under nonreducing conditions, ELISA, and immunohistochemistry of formalin-fixed paraffin-embedded tissue.

Applications Tested:

• Flow cytometric analysis of human peripheral blood cells (≤0.5 µg/test).
• Immunohistochemistry on formalin-fixed paraffin-embedded human tissue using low pH antigen retrieval (≤40 µg/mL).
  Careful titration is recommended for optimal assay performance.

Target Information

The CD93 protein is a cell-surface glycoprotein and type I membrane protein originally identified as a myeloid cell-specific marker. It is involved in intercellular adhesion and clearance of apoptotic cells. The intracellular cytoplasmic tail interacts with moesin, linking transmembrane proteins to the cytoskeleton and contributing to cytoskeletal remodeling.
