Product Specific Information
This 22F6 monoclonal antibody reacts with human, mouse, and canine Helios.
Helios is a zinc-finger transcription factor similar to Ikaros, expressed in hematopoietic stem cells of embryos and adult mice.
Highest expression occurs in regulatory T cells, while mature B cells, dendritic cells, and myeloid cells do not express Helios.
Helios can form homodimers or heterodimers with other Ikaros family members to regulate transcription.
Eight isoforms exist, some with dominant-negative functions upregulated in lymphoid malignancies such as T-acute lymphoblastic leukemia.
22F6 has been reported to cross-react with rat Helios.
Applications Reported:
- Intracellular staining followed by flow cytometric analysis.
Applications Tested:
- Pre-diluted and tested using mouse lymph node cells with Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
- Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume).
- Cell number range: 10⁵–10⁸ cells/test.
Fluorophore Information:
- Brilliant Ultra Violet™ 395 (BUV395) emits at 395 nm.
- For use on cytometers equipped with an ultraviolet (355 nm) laser.
- Verify instrument compatibility before use.
Staining Panel Recommendation:
When using multiple polymer dye-conjugated antibodies (e.g., Brilliant Violet™, Brilliant Ultra Violet™), use:
- Super Bright Complete Staining Buffer (Product #SB-4401-42)
- or Brilliant Stain Buffer™ (Product #00-4409-75)
to minimize non-specific polymer interactions.

Target Information
The Helios gene encodes a member of the Ikaros family of zinc-finger proteins.
These hematopoietic-specific transcription factors (Ikaros, Aiolos, Helios) regulate lymphocyte development.
Helios forms homo- or hetero-dimers with other Ikaros family members and functions primarily in early hematopoietic development.
Multiple transcript variants exist; biological validity of some remains uncertain.
