Product Specific Information

Description:
The 22F6 monoclonal antibody reacts with human, mouse, and canine Helios, a zinc-finger transcription factor related to Ikaros. Helios is expressed in hematopoietic stem cells, with the highest expression in regulatory T cells, but not in mature B cells, dendritic cells, or myeloid cells. It can form homo- and heterodimers with other Ikaros family proteins to regulate transcription. Certain isoforms have dominant-negative functions and are upregulated in lymphoid malignancies such as T-acute lymphoblastic leukemia.
22F6 has also been reported to cross-react with rat Helios.

Applications Reported:
For intracellular staining followed by flow cytometric analysis.

Applications Tested:
Prediluted and tested for intracellular staining of mouse lymph node cells using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00). Recommended use at 5 µL (0.125 µg) per test in a 100 µL final volume. Suitable for 10⁵–10⁸ cells/test.

Fluorochrome Information:
Brilliant Ultra Violet™ 395 (BUV395) emits at 395 nm and is compatible with cytometers equipped with a 355 nm UV laser.

Staining Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific interactions.

Target Information

This gene encodes a member of the Ikaros family of zinc-finger proteins, which includes Ikaros, Aiolos, and Helios. These transcription factors are hematopoietic-specific and regulate lymphocyte development. The protein can form homo- or heterodimers with other Ikaros family members and functions mainly in early hematopoietic development. Multiple transcript variants exist, though not all have confirmed biological validity.
