Target Information
STAT4 was originally identified using degenerate primers complementary to conserved sequences of other STAT proteins. The STAT4 protein shares similarity with STAT1 (52%) and STAT3 (47%). Functionally, STAT4 is tyrosine phosphorylated by Jak1 or Jak2 and forms homo- or heterodimers with other STAT family members. Tyrosine-phosphorylated STAT4 binds the IFN-gamma activated site (GAS), and serine phosphorylation is required for maximal transcriptional activity.
STAT4 expression is mainly found in the thymus, spleen, and testis. It is activated by interleukin-12 (IL-12), which is essential for T-cell independent induction of IFN-gamma and development of Th1 responses against intracellular pathogens. STAT4-deficient mice show impaired IL-12-induced Th1 development and enhanced Th2 differentiation. In mice, IFN-α/β activation of STAT4 is required for IFN-γ production during viral infection.
