Product Specific Information

Description:
The 2D10.4 monoclonal antibody reacts with human CD80 (B7-1), a 60 kDa member of the immunoglobulin superfamily. CD80 is expressed by activated B cells, macrophages, and dendritic cells, and also by activated T cells. CD80 binds to T cell surface antigens CD28 and CD152 (CTLA-4), playing a critical role in T-B cell communication and activation.

Applications Reported:
The 2D10.4 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use ≤ 1 µg per test (in 100 µL final volume). Optimal antibody titration is advised depending on cell number (10⁵–10⁸ cells/test).

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD80 (B7-1) and CD86 (B7-2) are ligands of the T cell costimulatory molecule CD28 and inhibitory receptor CTLA-4 (CD152). Both are expressed on professional antigen-presenting cells and are essential for T cell activation. CD80 expression is induced on activated B cells, EBV-transformed B cell lines, Burkitt’s lymphoma cells, T cells, and monocytes.
B7-1 provides regulatory signals for T lymphocytes through binding to CD28 and CTLA4.
CD80 dysfunction has been linked to diseases such as gallbladder squamous cell carcinoma and myocarditis.
