Product Specific Information

The 4SM15 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ or CD8αα dimers. CD8 binds to MHC class I and, through association with protein tyrosine kinase p56lck, plays a role in T cell development and activation.

Recent studies indicate that 4SM15 recognizes a non-CD8a epitope in neural tissue; therefore, it is not recommended for staining mouse neural tissue.
For detection in FFPE mouse neural tissue, use Anti-Mouse CD8a Purified, clone 4SM16 (Product #14-0195).
For frozen mouse tissue IHC, use Anti-Mouse CD8a Purified, clone 53-6.7 (Product #14-0081-82).

Applications Reported: Flow cytometry, microscopy, and IHC of formalin-fixed paraffin-embedded (FFPE) sections.
Applications Tested:

• IHC (FFPE mouse tissue): ≤10 µg/mL
• Flow cytometry (mouse splenocytes): ≤0.25 µg/test (100 µL final volume; 10⁵–10⁸ cells/test)
  Careful titration is recommended for optimal performance.

Target Information

CD8 is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It plays a key role in signal transduction, T cell differentiation, and activation.

• Expressed primarily on T cells as CD8αβ heterodimers, functioning as co-receptors for MHC class I molecules.
• CD8α homodimers are also found on subsets of T cells, NK cells, monocytes, and dendritic cells.
• Engagement of MHC-I/peptide complexes triggers recruitment of Lck, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation.
• Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
