The KGP130 monoclonal antibody reacts with mouse gp130 (CD130), a 130 kD type I transmembrane glycoprotein with a 597 amino acid extracellular domain, a single transmembrane domain, and a 277 amino acid cytoplasmic domain. gp130 is a subunit of several heterodimeric cell-surface receptors, including IL-6, IL-11, IL-27, Oncostatin M, and LIF receptors. A soluble form of gp130 can inhibit IL-6 signaling.

gp130 is mainly expressed on T cells, monocytes, endothelial cells, activated B cells, and plasma cells, with lower expression on most leukocytes and epithelial cells. Upon ligand binding, gp130 becomes tyrosine phosphorylated, activating signaling pathways such as PI3 kinase, Ras-MAPK, and STAT.

Preincubation of mouse splenic T cells with IL-6 can block KGP130 binding, suggesting the KGP130 epitope overlaps the IL-6 interaction site. No reactivity with human gp130 has been observed.

Target Information

The gene encodes a signal transducer shared by multiple cytokines, including IL-6, CNTF, LIF, and OSM. It functions as part of the cytokine receptor complex, activated upon cytokine binding. The viral IL-6 (vIL6) protein from Kaposi sarcoma-associated herpesvirus can directly activate this protein. Mouse knockout studies indicate a role in regulating myocyte apoptosis. Alternative splicing produces distinct isoforms.
