Product Specific Information

The YB5.B8 monoclonal antibody reacts with human CD117, also known as c-Kit, Steel factor receptor and stem cell factor receptor. This 145 kDa tyrosine kinase receptor is expressed by hematopoietic progenitor cells and mast cells. Interaction of c-Kit and Steel factor promotes proliferation and differentiation of hematopoietic progenitor cells and mast cell differentiation. CD117 is also expressed by melanocytes and plays a role in their activation.

For staining HSCs from peripheral blood, clone 104D2 is recommended. Refer to Product #17-1178.

Applications Tested: Pre-titrated and tested on TF-1 (human erythroleukemia) cell line at 5 µL (0.5 µg)/test.

Target Information

KIT (c-KIT) is a proto-oncogene and a type 3 transmembrane receptor for MGF (mast cell growth factor, stem cell factor). KIT regulates growth and activation of hematopoietic and non-hematopoietic cells. Mutations in KIT are associated with gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism. Deregulation of KIT receptor TK (e.g., mutation D816V) is a therapeutic target in neoplastic mast cell growth.

c-Kit is expressed in hematopoietic stem cells, germ cells, mast cells, and gastrointestinal tract cajal cells. Upon SCF binding, c-Kit dimerizes, leading to receptor activation and autophosphorylation, enabling downstream signaling pathways (Ras/ERK, PI3K, PLC-γ, JAK/STAT). c-Kit activation is essential for hematopoiesis, stem cell maintenance, and gametogenesis.
