Product Specific Information

The monoclonal antibody 3E4 reacts with human and mouse interferon regulatory factor 4 (IRF4), a 52 kDa transcription factor involved in B cell, T cell, and macrophage function.

• In B cells, IRF4 is highly expressed in mature plasma cells and regulates differentiation, interacting with PU.1 and controlling transcription of B cell–specific genes such as Prdm1 (Blimp1).
• In T cells, IRF4 contributes to Treg, Th2, Th9, and Th17 cell development and function. It is upregulated upon activation and induces Blimp1 and IL-10 expression.
• IRF4 also plays a role in macrophage polarization and is upregulated in several hematologic malignancies.

Applications Reported: Western blotting.
Applications Tested: Western blot on reduced splenocyte lysate cultured with plate-bound Anti-Mouse CD3 and soluble Anti-Mouse CD28. Recommended ≤ 5 µg/mL; titrate for optimal performance.

Target Information

The IRF (interferon regulatory factor) family of transcription factors share a tryptophan pentad repeat DNA-binding domain. They regulate interferon responses and interferon-inducible genes.
IRF4 is lymphocyte-specific and negatively regulates TLR signaling, essential for innate and adaptive immunity.
A chromosomal translocation t(6;14)(p25;q32) involving IRF4 and IgH may cause multiple myeloma.
Alternatively spliced transcript variants have been identified.
