Specificity Comments:
Beta-catenin associates with the cytoplasmic portion of E-cadherin, essential for its adhesion function. In normal tissues, beta-catenin localizes to epithelial cell membranes. In breast ductal neoplasia, it is membrane-localized, while in lobular neoplasia, it redistributes throughout the cytoplasm, showing diffuse staining. Immunostaining of beta-catenin and E-cadherin aids in distinguishing ductal and lobular neoplasms, including DCIS and lobular carcinoma. Some rectal and gastric adenocarcinomas exhibit diffuse cytoplasmic beta-catenin staining, similar to lobular breast carcinomas.

Target Information

Beta-catenin is an adherens junction (AJ) protein involved in cell-cell adhesion and epithelial layer maintenance. It interacts with E-cadherin and alpha-catenin, linking to the cortical actin cytoskeleton. Beta-catenin also binds to complexes containing adenomatous polyposis coli protein and microtubules, engaging signaling pathways such as Wnt/Wingless. It plays roles in morphogenesis and is expressed in various hair follicle cell types. Pathological roles have been observed in pilomatrixoma, medulloblastoma, colorectal, ovarian, and other cancers. In the nucleus, beta-catenin coactivates Lef/Tcf transcription factors, promoting expression of cyclin D and c-myc, key regulators of cell proliferation and tumorigenesis.
