Target Information
Beta-catenin is an adherens junction (AJ) protein originally identified as a component of cell-cell adhesion structures. AJs are essential for the formation and maintenance of epithelial cell layers by regulating cell growth and adhesion. Beta-catenin interacts with the cytoplasmic domain of E-cadherin and links it to alpha-catenin, facilitating anchorage of the E-cadherin complex to the actin cytoskeleton. It also associates with complexes containing adenomatous polyposis coli protein and microtubules, participating in signaling pathways such as tyrosine kinases, phosphatases, and Wnt/Wingless.
Beta-catenin is expressed in various hair follicle cell types and plays roles in morphogenesis and tumorigenesis. Dysregulation has been associated with pilomatrixoma, medulloblastoma, colorectal and ovarian cancers, and other solid tumors. In the nucleus, beta-catenin co-activates Lef/Tcf transcription factors, promoting transcription of genes like cyclin D and c-myc that drive cell proliferation.
