Product Specific Information
The E11/19 monoclonal antibody binds to phosphoserine 349 corresponding to the C-terminal 22 amino acid residues of human CCR5. CCR5 acts as a receptor for inflammatory CC-chemokines such as CCL3 (MIP-1α), CCL4 (MIP-1β), and RANTES.
Target Information
Human immunodeficiency virus (HIV) and related viruses require coreceptors in addition to CD4 to infect target cells. CCR5, along with CXCR4, CCR3, CCR2b, and CCR8, are chemokine receptors identified as HIV coreceptors. CCR5 (CC-CKR-5) is a principal coreceptor for macrophage- and dual-tropic HIV-1 strains, facilitating fusion and entry into human white blood cells. CCR5 is essential for infection by HIV-1, HIV-2, and SIV. Beta-chemokines RANTES, MIP-1α, and MIP-1β are natural ligands for CCR5 and can prevent M-tropic HIV-1 infection. CCR5 interacts with the CD4-gp120 complex of HIV, leading to membrane fusion and viral entry. CCR5 mRNA is expressed in lymphoid organs and monocytes.
