Specificity: The 32-G1 antibody recognizes C5aR with phosphorylated serine 334. It specifically reacts with a C5aR peptide containing phosphoserine 334 and with C5aRs isolated from C5a-stimulated, transfected RBL-C5aR cells.
It does not recognize a receptor mutant (C5aR-S--) where all six C-terminal serine phosphorylation sites are replaced with alanine.
Antibody 32-G1 specifically reacts with C5aR phosphoserine 334, independent of neighboring serine residues’ phosphorylation state.
In Bt2cAMP-treated U937 cells, 32-G1 reacts specifically with ligand-activated C5aR at endogenous expression levels but only weakly with unstimulated cells.

Target Information

CD88 (C5aR1) is a C5a Anaphylatoxin Receptor mediating effects of potent chemotactic agents.
C5AR1 is implicated in asthma and expressed in myeloid blood cells, brain, liver, lung, spleen, heart, kidney, and intestinal tract.
CD88 binds the inflammatory peptide anaphylatoxin C5a, stimulating chemotaxis, granule enzyme release, and superoxide production.
It is part of the classical complement cascade and may contribute to tissue injury related to ischemia or sepsis.
Associated diseases include Mannose-Binding Lectin Deficiency and Systemic Lupus Erythematosus.
